> The trial showed significant results for both progression-free and overall survival. Genmab plans to discuss next steps with regulators; its shares rose 9.48% at the Oct. 23 close.

Genmab said Oct. 23 its investigational antibody-drug conjugate rinatabart sesutecan (Rina-S) met its primary progression-free survival and key secondary overall-survival goals in a Phase 3 trial of platinum-resistant ovarian cancer. The topline results favored Rina-S over investigator-selected chemotherapy.

In the 584-patient intent-to-treat population, Genmab reported a 71% reduction in the risk of disease progression or death, with a hazard ratio of 0.29 (95% confidence interval, 0.23-0.37; P<0.0001). In the 492-patient primary analysis set, the reported risk reduction was 67%, with a hazard ratio of 0.33 (95% confidence interval, 0.26-0.42; P<0.0001).

Overall-survival results were statistically significant in both groups. The hazard ratio was 0.43 in the intent-to-treat population (95% confidence interval, 0.31-0.59; P<0.0001) and 0.44 in the primary analysis set (95% confidence interval, 0.31-0.63; P<0.0001). Genmab did not provide median survival figures in its announcement.

Enrollment did not depend on patients’ FRα expression or whether they had previously received mirvetuximab. Genmab said the observed safety profile was consistent with Rina-S’s known safety profile, with no new safety signals or unexpected safety concerns. Rina-S targets FRα and carries exatecan. The chemotherapy options included paclitaxel, topotecan, pegylated liposomal doxorubicin and gemcitabine.

Rina-S remains investigational. Genmab plans to discuss next steps with global regulators and submit the results for presentation at a future medical meeting. The announcement did not give a meeting date. The Phase 3 results follow [earlier coverage of Rina-S combination safety results](<https://www.tokenpost.com/news/business/21312>).

At the 4 p.m. ET (20:00 UTC) close on Oct. 23, Genmab shares were $43.79, up 9.48% from $40 on Oct. 22. Corcept shares closed at $85.86, down 20.13% from $107.50. These close-to-close changes do not show how much the trial announcement contributed to either stock’s move.

The Food and Drug Administration (FDA) approved Corcept’s Lifyorli (relacorilant) with nab-paclitaxel on March 25 for eligible adults with platinum-resistant ovarian, fallopian tube or primary peritoneal cancer after one to three prior systemic treatment regimens, including at least one with bevacizumab. Lifyorli and Rina-S are separate treatments.
